全反式维甲酸对抗肿瘤药物促进HL-60细胞凋亡作用的影响

摘要 目的观察诱导分化剂对抗肿瘤药物促进HL60细胞凋亡的影响。方法采用苏木精染色、流式细胞仪及DNA电泳的方法,研究全反式维甲酸(ATRA)分别与阿糖胞苷、长春新碱、鬼臼乙叉甙、柔红霉素4种抗肿瘤药协同作用后HL60细胞凋亡的发生情况。结果当先以ATRA作用HL60细胞3天后再分别给予抗肿瘤药物,发现各个药物引起的凋亡细胞的数量减少60%以上。反之,先以抗肿瘤药作用后再以ATRA处理HL60细胞,各个药物可使凋亡细胞的数量增加15%以上。结论ATRA对某些抗肿瘤药的促HL60细胞凋亡作用可能有一定影响。 Objective To observe the effects of differentiating drugs on the response of HL60 cells to apoptosis induced by antitumor drugs. Methods The responses of HL60 cells to apoptosis induced by AraC, VCR, VP16 and DNR in combined with ATRA were studied by detection of apoptotic cells, and hematoxylin staining, flow cytometry and DNA electrophoresis were used to assay apoptosis. Results The number of apoptotic cells decreased by over 60% with administration of ATRA three days before treating HL60 cells with four antitumor drugs respectively, while the apoptosis increased by over 15% with administration of drugs in reverse sequence. Conclusion The results of this study suggest that ATRA might probably affect the susceptibility to apoptosis of HL60 cells induced by AraC, VCR, VP16 and DNR.
