摘要
目的:研究镉导致核苷酸切除修复基因XPC在HEK293细胞中的表达变化,探讨XPC基因在镉导致细胞DNA突变种的作用。方法:离体培养的人胚肾293细胞以20umol/L(低剂量组)和40umol/L(高剂量组)分别氯化镉处理24小时,Real-timePCR检测处理前后XPC的表达变化情况。结果:两组中,氯化镉处理前后XPC的表达明显升高(P<0.01);高剂量组中XPC的表达与低剂量组中XPC的表达没有明显差别(P>0.05)。结论:高表达的XPC、XPF在镉导致的DNA突变中起着重要的修复作用,核苷酸切除修复机制对镉造成的DNA损伤具有一定的修复作用。
Objective: Little is known now about the function of NER in the HEK293 cell induced by the cadmium, we investigated the mRNA expression of Xeroderma Pigmenosum group C in HEK293 cell induced by cadmium in different concelttration. Method: Transformed human embryonic kidney 293 cell line were incubated with 40μ mol/L or 80μ mol/L CdCl 2 for 24 hours Expression levels of xpc were detected with the real-time polymerase chain reaction. Results: The xpc gene was frequently overexpressed in HEK293 cell induced by cadmium compared to normal HKE293 cell. Conclusion: Our findings suggest that overexpression of xpc gene involved in the early steps of NER process and could be related to the well recognized resistance of cadmium in some degree.
