摘要
目的:观察岩黄连总碱对体外培养人舌鳞癌Tca8113细胞增殖抑制及诱导凋亡作用。方法:MTT法检测不同浓度的岩黄连总碱分别处理体外培养的Tca8113细胞24、48和72 h后的细胞增殖抑制情况,Hochest33258染色及DNA ladder检测细胞凋亡,并用脱氧核苷酸转移酶介导的dUTP缺口末端标记法(TUNEL法)原位检测细胞凋亡及细胞凋亡率。结果:岩黄连总碱对Tca8113细胞增殖具有明显抑制作用(P<0.01),且呈现出一定的时间和浓度依赖性;Hochest33258检测发现岩黄连总碱作用48 h后可见凋亡细胞;DNA ladder检测可见凋亡特有的180~200 bp整数倍DNA条带;TUNEL结果显示岩黄连总碱处理组细胞凋亡率呈时间和浓度依赖性。结论:岩黄连总碱能抑制Tca8113细胞增殖,呈时间和浓度依赖性;同时能诱导Tca8113细胞凋亡,且诱导凋亡的作用也呈时间和浓度依赖性。
Objective: To investigate the effects of Corydalis saxicola Bunting total alkaloids on cell proliferation and apoptosis of human tongue squamous carcinoma Tea8113 cells in vitro. Methods: Tca8113 were treated by different concentrations of Corydalis saxicola Bunting total alkaloids for 24, 48, 72 h respectively. Cell proliferation inhibition was assayed with MTT. Hochest33258 staining and DNA ladder were used to detect their apoptosis. And TUNEL was used to detect apoptosis in sitn and apoptosis rate. Results: Cell proliferation was significantly inhibited by Corydalis saxieola Bunting total alkaloids in a time- and dose-dependent manner (P〈0.01). After exposure to Corydalis saxicola Bunting total alkaloids for 48 h, the apoptotic cells could be detected by Hochest33258 staining, and integer multiples of 180-200 bp DNA apoptosis-specific fragments could be detected by DNA ladder. The results of TUNEL showed the apoptosis rate changed in a time- and dose-dependent manner. Conclusion: Corydalis saxicola Bunting total alkaloids can inhibit the proliferation and induce the apoptosis of Tea8113 cells in a time- and dose-dependent manner.
