摘要
目的构建并制备血管内皮生长因子(VEGF)重组腺病毒,观察VEGF基因转染对骨髓基质干细胞(MSCs)的增殖和诱导为神经细胞能力的影响。方法将VEGF的基因亚克隆至穿梭载体;使用Padeasy-1腺病毒系统制备包含VEGF基因的腺病毒。得到的腺病毒感染MSCs,使用无血清诱导法使其向神经元细胞分化。鉴定诱导分化结果,评价VEGF基因转染对MSCs向神经元细胞诱导分化的影响。结果成功制备出编码VEGF的腺病毒,病毒滴度为1×1010v.g./L,以MO I=100感染MSCs,无血清诱导条件下VEGF基因转染的MSCs向神经细胞分化的结果与未转染的MSCs无明显差异。结论 VEGF基因转染对MSCs向神经细胞诱导分化无明显影响,为下一步应用于临床奠定了基础。
Objective To construct adenovirus encoding vascular endothelial growth factor (VEGF) and evaluate the effect of VEGF transfection on proliferation and differentiation of marrow stroma cells (MSCs) into neuronal ceils. Methods The adenovirus pAd-VEGF encoding VEGF was established by the p-shuttle and Padeasy- 1 system. MSCs were acquired and cultured by svandard method. Differentiation of MSCs into neuronal cells was valued after MSCs were transfected by VEGF at MOI = 100. Results The recombinant adenovirus vector was successfully established with tilter of 1×10^10v. g./L. Compared with control, there was no significant difference of differentiation ability of MSCs into neuronal cells after transfection of VEGF. Conclusion VEGF gene modification has not significant effect on differentiation ability of MSCs into neuronal cells.
