草菇SCAR遗传标记建立及其杂种鉴定应用 被引量:17

摘要 草菇菌丝细胞多核且无锁状联合,缺乏明显的形态标记,遗传育种存在难题。采用SRAP法筛选出亲本单26和亲本单28的遗传差异条带4条,经克隆测序,并根据序列设计的引物优化扩增温度条件后,成功转化成SCAR遗传标记,其中SCAR1与SCAR4为亲单26所特有,SCAR2与SCAR3为亲单28所特有。应用这4个遗传标记,成功地鉴定了菌株2628的杂种性,并将应用于杂种后代单孢分离物的遗传分析等研究,为揭示草菇的性遗传模式提供有效的手段。 There are multi-nucleus in hypal cells and no clamp-connections in morphology onVolvariella volvacea . That makes its difficulty to cross breeding. In this study, using SRAP method for screen out different genetic bands between single-spore strain 26 and strain 28, and optimizing temperature conditions of PCR primers designed on sequence of bands, finally, we got four different SCAR genetic markers, which SCAR1 and SCAR4 belong to strain 26 specially, SCAR2 and SCAR3 belong to strain 28 specially. This four SCAR genetic markers have successful identified the hybrid nature of the strains 2628 and will be applied to genetic analysis of hybrid offspring. It reveals the nature of the genetic model of straw mushroom and provides an effective means.
