摘要
目的探讨内异康复片对子宫内膜异位症(EMT)大鼠异位和在位内膜血管形成的不同影响,揭示内异康复片治疗EMT的作用机理。方法建立大鼠EMT模型,随机分为内异康复片高、中剂量组、丹那唑组、丹莪妇康煎膏组、模型组和空白组,采用免疫组化SP法染色,半定量检测血管内皮生长因子(VEGF)、内皮抑制素(ENS)在异位和在位内膜中的表达,并通过CD31标记异位子宫内膜血管内皮细胞,测定其微血管密度(MVD)。结果内异康复片高、中剂量组均能显著降低EMT大鼠异位内膜VEGF的表达(P<0.01),升高ENS的表达(P<0.01),并能显著降低EMT大鼠异位内膜血管内皮生长因子/内皮抑制素(V/E)值、MVD(P<0.01)和微血管数目(P<0.05),而对大鼠在位内膜V/E值、MVD及微血管数目的表达与空白组比较差异无统计学意义(P>0.05)。结论内异康复片可能通过对血管形成过程的影响,抑制异位内膜组织血管的生成,促进异位病灶消退,同时使血管形成刺激因子和抑制因子之间相互平衡,但不抑制在位内膜正常血管增生的生理过程。内异康复片可降低异位子宫内膜微血管密度(MVD),可能与异位内膜组织血管生成受到抑制有关。
Objective To discover the mechanism of Neiyikangfu Tablets in treating endometriosis by studying the influence to hemangiogenic factors on rats' in utero and ectopic endometrium. Methods Established endometriosis models of rats and distributed them randomly into high dose and medium dose group of Neiyikangfu Tablets,Danazol group,Danefukang condensed decoction group,model group and blank group. Dyeing with immuno-histochemical SP,semiquantitatively detected the expression of VEGF and ENS in utero and ectopic endometrium,and signed ectopic endometrial vascular endotheliocyte by CD31 to detect MVD.Results The expression of VEGF,V/E,MVD and number of micro vascular of ectopic endometrium was low and ENS was high in high and medium dose Neiyikangfu Tablets groups (P0.01).But there was no difference in V/E,MVD and number of micro vascular between in utero endometrium group and blank group(P0.05). Conclusions Maybe Neiyikangfu Tablets can inhibit the angiogenesis process of ectopic endometrium and improve the extinction of ectopic endometrium,help keeping the balance of stimulating and inhibiting factors of angiogenesis,but can't inhibit the physiologic angiogenesis process of in utero endometrium. It can reduce MVD of ectopic endometrium maybe by inhibiting the angiogenesis of ectopic endometrium.
