条斑紫菜多糖对人肝癌Bel7402抗肿瘤作用的初步研究 被引量:9

摘要 目的:研究条斑紫菜多糖对人肝癌Bel7402体外抗肿瘤作用。方法:采用MTT法观察条斑紫菜多糖体外对肝癌Bel7402的增殖抑制作用;荧光显微镜观察不同浓度条斑紫菜多糖诱导Bel7402细胞凋亡作用情况,RT-PCR检测Survivin、Bax基因表达量的变化。结果:条斑紫菜多糖对人肝癌Bel7402细胞有显著地增殖抑制作用,能诱导Bel7402细胞凋亡,并且能上调Bax、降低Survivin表达。结论:条斑紫菜多糖体外有一定的抗肿瘤作用,并能促进Bel7402细胞凋亡,其机制可能与激活Bax、抑制Survivin表达有关。 [Objective] To observe the anti-tumor activity of Porphyra yezoensis polysaccharide on Be17402 cells in vitro. [Method] In vitro, the inhibitory effects of Porphyra yezoeasis polysaccharide on proliferation of Be17402 cells were measured by MTF colorimetric assay. Be17402 cells were treated with various doses of Porphyra yezoensis polysaccharide. Apoptosis of Be17402 cells were observed by fluorescence microscope. The levels of survivin and bax gene expression were detected by RT-PCR. [Result] The studies demonstrated that Porphyra yezoensis polysaccharide inhibited proliferation of Be17402 ceils. There was obviously apoptosis-inducing effect on Be17402 cells and it could effectively decrease the level of survivin gene expression and improve the level of bax gene expression. [Conclusion] Porphyra yezoensis polysaccharide had anti-tumor effect on Be17402 cells and could induce apoptosis of Be17402 cells probably through activating survivin gene expression and inhibiting bax gene expression.
