不卵裂受精卵微管蛋白/纺锤体和核/染色体形态观察 被引量:2

Spindle/microtubule and nuclear/chromosome abnormalities in arrested human zygotes during procedure of IVF/ICSI

摘要 目的:观察在不卵裂受精卵中核/染色体和微管蛋白/纺锤体的形态,探讨在有丝分裂过程中染色体和纺锤体形态异常是否参与了受精卵的不卵裂。方法:采用免疫荧光法对核/染色体和微管蛋白/纺锤体进行染色,并用激光共聚焦显微镜观察它们的形态。结果:在不卵裂受精卵中,有37.50%的受精卵处于间期,受精后不能进入有丝分裂期;有62.50%的受精卵处于有丝分裂期,微管蛋白形态异常。结论:在正常受精后,受精卵在细胞周期的转换及微管蛋白/纺锤体的形态上存在异常,这可能是导致受精卵不卵裂的原因之一。 Objective:To investigate the nuclear/chromosome and microtubule/spindle abnormalities in arrested human zygotes in vitro.Methods:To investigate this,fluorescence and confocal laser scanning microscopy were used,following immunolabelling with antibodies specific for α-tubulin,combined with DNA fluorochrome.Results:62.50% of the arrested human zygotes had abnormalities in the organization of microtubule,and 37.50% of the arrested human zygotes were in the interphase of the cell cycle.Conclusion:We propose that interphase/metaphase transition arrest,microtubule organization and spindle abnormalities cause human one-cell embryo development arrest.
