摘要
利用硝基化反应,探索建立抗糖尿病体外快速筛选模型。本研究通过形成的3-硝基酪氨酸(3-n itrotysine,3-NT)在紫外425 nm吸收强度的变化,对民间草药翻白草治疗糖尿病的验方进行药效活性成分。根据翻白草不同提取物对硝基化反应的抑制作用,建立了简便可靠的抗糖尿病体外筛选和快速评价方法。实验基于从翻白草所得的甲醇、乙醇不同浓度和总黄酮等不同部位的提取物,经体外硝基化反应评价,筛选出翻白草的甲醇萃取物、95%乙醇抽提物、总黄酮、聚酰胺层析的40%乙醇洗提物和85%乙醇洗提物为有效抗糖尿病部位,均有明显的抑制蛋白硝化的作用。该研究通过优化外硝基化反应最佳条件,进而建立的体外硝基化反应模型,揭示了翻白草提取物含有抑制硝化的物质即为该植物的抗糖尿病成分。因此,抑制蛋白硝基化作用可以考虑作为体外抗糖尿病的有效手段,并且有望完善成为体外抗糖尿病药物快速筛选的简便方法和模型之一。
Using nitration rapid screening the anti-diabetic model in vitro was established.Based on the absorption intensity of 3-nitrotyrosine at 425 nm,folk prescription Potentilla Discolor.Bunge was studied on its active ingredients.The study method adopted anti-free radical model in vitro according to free radical theory aboat diabete.According to the anti-nitrations of different extracts,a method to screen active ingredients of anti-diabetes mellitus rapidly was esteblished in vitro.Mathanol extract,different concentration ethanol extracts and totol flavonoids were evaluated by this model.There was supiorer inhibition activity in fraction as follows:mathanol extract,95% ethanol extract,total flavonoids,40% ethanol eluteds and 85% ethanol eluteds from the column chromatography of polyamide.The results showed that there are anti-diabitic substances in Potentilla Discolor.Bunge which provides the basis study for chemical separating.Therefore,inhibitory protein nitration reaction can be considered as an effective means of anti-diabetes in vitro.Besides,it can be refined into one of simple methods and models to rapidly screen anti-diabatic drug in future.
