摘要
目的:通过观察黄芪对自然衰老小鼠皮肤组织结构及线粒体T-ATP酶活性的影响,探讨黄芪延缓自然衰老小鼠皮肤衰老的可能机制。方法:选用健康昆明种小鼠分青年对照组(A)、老龄空白对照组(B)、黄芪低剂量组(C)、黄芪中剂量组(D)、黄芪高剂量组(E)和维生素EC组(F),采用电镜观察小鼠皮肤组织超微结构的改变,以生化分析方法测定皮肤组织中线粒体总ATP酶活性。结果:自然衰老小鼠皮肤组织细胞氧化损伤,线粒体肿胀,出现空泡化。与青年对照组相比较,老龄空白对照组(P<0.01)、黄芪低剂量组(P<0.01)、黄芪中剂量组(P<0.01)、黄芪高剂量组(P<0.01)、维生素EC组(P<0.01)线粒体ATP酶活性均明显降低。黄芪低剂量组(P<0.01)、黄芪中剂量组(P<0.01)、黄芪高剂量组(P<0.01)、维生素EC组(P<0.01)均可提高皮肤组织中线粒体ATP酶活性。结论:自然衰老小鼠的皮肤组织中有明显的氧化损伤,胶原蛋白减少,线粒体ATP酶活性明显降低。黄芪对皮肤组织成纤维细胞的细胞核、内质网、线粒体等损伤具有保护作用。其发挥延缓皮肤衰老的作用机制可能是通过清除自由基,提高皮肤中线粒体ATP酶活性,提高线粒体呼吸功能,促进成纤维细胞合成胶原,提高机体抗氧化能力密切相关。
Objective:To study mtDNA default and change of mitochondrial ATPase of the skin tissue in aging mice from the intervention by Radix Astragali.So as to explore the mechanism of staying senescence of skin by traditional Chinese herb Radix Astragali.Methods:72 healthy Kunming mice were selected and divided as young control grou(pA),blank old age control grou(pB),low dosage group of Radix Astragal(iC),medium dosage group of Radix Astragali(D),high dosage group of Radix Astragali(E)and Activated group of old age(F).Observe the change of ultrastructure in skin tissue of mice by electro-microscope and analysis the activity of SOD and ATP enzyme of mitochondria in skin tissue by biochemistry technique.Results:There are oxidation damage in skin tissue of the natural senescence mice.Contrasting with young control group,the activity of ATP enzyme of mitochondria evidently depressed(separately B group is P〈0.01,C group is P〈0.01,D group is P〈0.01,E group is P〈0.01,F group is P〈0.01).The low,medium and high dosage groups of Radix Astragali and Activated group of old age all can heighten the activity of ATP enzyme of mitochondria in skin tissue of mice(separately C group is P〈0.05,D group is P〈0.05,E group is P〈0.05,F group is P〈0.01).Conclusion:There are oxidation damage evidently and Collagen protein reducing,the activities of ATP enzyme of Mitochondria reducing evidently in skin tissue of the natural senescence mice.Traditional Chinese herb Radix Astragali may staves the senescence action by cleaning out the free Radical,enhancing breath function of Mitochondria and the activity of ATP enzyme,accelerating producing collagen from Fbs cell,improving the ability of antioxidant.
