疏肝导浊方干预家兔肝脏脂肪变的实验研究

Experimental Study on Liver Steatosis Interfered with Shugandaozhuo Complex Prescription in Rabbits

目的:观察疏肝导浊方对家兔实验性肝脏脂肪变的干预作用。方法:42只家兔随机分为正常组、模型组、疏肝导浊方低、中、高剂量组和饮食治疗组,每组7只。正常组家兔以普通饲料喂养,按10ml/kg体重给予蒸馏水灌胃;模型组家兔予高脂饲料喂养12周后处死;疏肝导浊方低、中、高剂量组家兔在喂饲高脂饲料12周后改为普通饲料,分别按1、2、3g/kg体重给予疏肝导浊方药液灌胃;饮食治疗组家兔在喂饲高脂饲料12周后改为普通饲料。于16周末心脏采血后处死各组家兔,制备肝组织石蜡切片,光镜下评估肝脏脂肪变性和炎症活动程度;分离血清,测定TC、TG、HDL-C、LDL-C、ALT和AST浓度。结果:与模型组和饮食组比较,疏肝导浊方中、高剂量组家兔血清TC、TG、LDL-C、ALT和AST显著降低(P<0.05或P<0.01),HDL-C显著增高(P<0.01);疏肝导浊方中、高剂量组家兔肝脂肪变性程度和炎症活动度积分明显低于模型组(P<0.05)。结论:疏肝导浊方能改善实验性脂肪肝家兔的血脂和肝功能指标,减轻肝组织脂肪变性程度和炎症反应,以中、高剂量的作用最显著。

Objective：To explore the effects of Shugandaozhuo complex prescription on steatosis in the liver in rabbits. Methods： Forty - two rabbits were ran into 4 groups. Normal group was breed common forage. Model group was breed high lipoid forage. Shugandaozhuo complex prescription group was intragastric administered with Chinese materia medica after 12 -week high lipoid forage（ including 3 subgroups with different dosis）. Placebo group was breed common forage after 12 - week high lipoid forage. The model group were sacrificed 12 weeks later and other groups were killed 16 weeks later. Blood samples were harvested before putting to death and liver tissue samples were harvested for histological examination. The levels of total cholesterol （ TC ）, triglyceride （ TG ）, high density lipoprotein - cholesterol （ HDL - C ）, low density lipoprotein -cholesterol（ LDL- C ）, alanine aminotransferase （ALT） and aspartate aminotransferase （AST）. Results：Compared with the model and placebo groups, the levels of TC,TG, LDL - C, ALT and AST were dramatically decreased in the medium and high dosis subgroups of the Shugandaozhuo complex prescription group（ P 〈 0.05 or P 〈 0.01 ） and HDL - C was increased （P〈 0. 01 ）;adipose degeneration and inflammation was degraded as compared to the model group. Conclusion： Shugandaozhuo complex prescription may ameliorate the adipose degeneration and inflammation in the liver of rabbits by regulating serum lipids and improving liver function.
