大鼠缺血再灌注ICAM-1表达与小动脉损伤关系的实验研究 被引量:1

Relationship between expression of intercellular cell adhesion molecule-1 (ICAM-1) and the ischemia-reperfusion injury of small artery in rats

摘要 目的探讨大鼠缺血再灌注(I/R)时细胞间黏附分子-1(intercelluar adhesion molecule-1,ICAM-1)表达与小动脉形态学改变的关系及意义。方法 70只Wistar大鼠随机分为3组:Ⅰ组:正常对照组(10只),Ⅱ组:缺血组(10只),Ⅲ组:I/R组(50只)。Ⅰ组由三通管放血立即处死。Ⅱ、Ⅲ组10min内放血至血压为40mmHg,并维持45min。Ⅱ组缺血45min后处死,Ⅲ组缺血45min后进行输血输液治疗,分别于再灌注1、3、6、12、24h时点处死。免疫组化染色观测ICAM-1在各组大鼠脑、肾、四肢肌肉的小动脉内皮和平滑肌细胞表达的阳性细胞数,并用光镜和透射电镜观察小动脉形态学改变。结果 (1)Ⅰ组脑、肾及四肢肌肉的小动脉ICAM-1不表达;与Ⅰ组比较,Ⅱ组ICAM-1在大鼠脑、肾及四肢肌肉的小动脉的表达变化差异无统计学意义(P>0.05);Ⅲ组再灌注1h脑、肾及四肢肌肉的小动脉ICAM-1表达上调(P<0.05),再灌注3h肾及四肢肌肉的小动脉ICAM-1表达达高峰,再灌注6h脑的小动脉ICAM-1表达达高峰,持续至24h;(2)光镜和电镜下Ⅱ组小动脉内皮和平滑肌轻微损伤,Ⅲ组损伤加重,以再灌注3h损伤最为显著。结论 ICAM-1在大鼠小动脉表达上调介导了I/R小动脉的损伤。 Objective To investigate the changes of intercellular adhesion molecule-1(ICAM-1)and its effects on the ischemia-reperfusion injury of small artery.Methods Seventy Wistar Rats were randomly divided into control group(group Ⅰ,n=10),ischemia group(group Ⅱ,n=10),and ischemia-reperfusion group(group Ⅲ,n=50).The expression of ICAM-1 in the small artery of brain,kedney and skeletal muscle was observed by the immunohistochemistry technique after ischemia for 45 min and reperfusion for 1,3,6,12,24 h.Results(1)Compared with group Ⅰ,the expression of ICAM-1 in the small artery of group Ⅱdid not change(P0.05),but that of group Ⅲ increased after reperfusion for 1 h,and lasted 24 h(P0.05).The expression of ICAM-1 in the small artery of kedney and skeletal muscle reached peak after reperfusion for 3 h,and that of brain reached peak after reperfusion for 6 h.(2)The injury degree of small artery of group Ⅲ was more severe than that of group Ⅱ.The most severe histological damage was found in group Ⅲ after reperfusion for 3 h.Conclusion Up regulation of ICAM-1 expression may play a role in the ischemia-reperfusion injury of small artery.
