白蛋白酶联免疫分析试剂盒的研制

Establishment of an Enzyme Linked Immunosorbent Assay for Albumin

采用竞争性ELISA法检测人尿中白蛋白(Albumin,Alb)的含量。将Alb抗体包被96孔微孔板,Alb与辣根过氧化物酶结合形成酶标记物,建立了白蛋白酶联免疫分析(Alb-ELISA)方法。结果显示,本方法分析灵敏度为0.28 mg/L;批内、批间变异系数分别为2.63%～5.28%和2.40%～4.26%;回收率为95.0%～106.4%;高浓度Alb样品系列倍比稀释后,测定值与稀释度呈线性相关,相关系数为r=0.997 4。本试剂盒操作简便、快速,适用于临床检测和科研应用。

A sensitive and specific ELISA for Albumin was established by using Albumin antibody coated on the microtiter plate,Albumin labeled horseradish peroxidase（HRP）.The sensitivity of the assay was 0.28 mg/L.The intra-and inter-assay CVs were 2.63%-5.28%and 2.40%-4.26% respectively.The analytical recovery was 95.0%-106.4%,the coefficient correlation was 0.9974.This method for measuring Alb was rapid,sensitive and convenient.It is suitable for clinical detection and research application.
