摘要
目的:探讨卵泡液中肿瘤坏死因子α(TNF-α)和γ干扰素(IFN-γ)与颗粒细胞凋亡的关系。方法:应用TUNEL方法测定卵丘颗粒细胞凋亡率,应用酶联免疫方法(ELISA)测定成熟卵泡液中TNF-α和IFN-γ的浓度,探讨其相关性。结果:妊娠组颗粒细胞凋亡率为12.25±4.84%,明显低于未妊娠组的15.96±7.04%(P<0.05);TNF-α浓度与颗粒细胞凋亡率的相关系数r=0.56,P<0.05,两者存在显著的正相关关系;IFN-γ浓度与颗粒细胞凋亡率的相关系数r=-0.57,P>0.05,两者不存在相关关系。结论:卵泡液中TNF-α浓度与颗粒细胞凋亡呈正相关,IFN-γ浓度与颗粒细胞凋亡无相关关系,颗粒细胞凋亡率高的患者不易获得妊娠。
Objective:To investigate the relationship between the concentration of tumour necrosis factor-α(TNF-α) and interferon-gamma(IFN-γ) in follicular fluid and granulosa cell apoptosis in IVF-ET treatment.Methods: TUNEL was used to analyze granulosa cell apoptosis.Enzyme linked immunosorbent assay(ELISA) wasused to analyze the concentrations of TNF-α and IFN-γ in mature follicular fluid.The IVF treatment outcome was closely followed up.The relationship between the concentration and granulosa cell apoptosis rate was discussed by statistical analysis.Results: The apoptosis rate of the granulosa cells(15.96±7.04%) in the patients without successful IVF-ET outcome was significantly higher than that in pregnant group(12.25±4.84%),P0.05.Correlation coefficient(r) of the TNF-α and the granulosa cells apoptosis rate was 0.56,which had a statistical significance(P0.05),while the correlation coefficient of the IFN-γ and the granulosa cell apoptosis rate was-0.57,which had no statistical significance(P0.05).Conclusion: Higher concentration of TNF-α will cause higher granulosa cell apoptosis rate,while IFN-γ has no effect on the granulosa cells apoptosis.Women received IVF treatment with higher granulosa cells apoptosis rate in mature follicular fluid is more difficult to get pregnancy.
