丹参素预处理对脑梗死大鼠血浆纤溶激活系统的影响 被引量:10

摘要 目的观察丹参素预处理对脑梗死大鼠血浆纤溶激活系统的影响。方法 40只雄性大鼠,随机分为假手术组、脑梗死对照组、低剂量[5mg/(kg·d)]丹参素组和高剂量[10mg/(kg·d)]丹参素组,每组10只。应用自体血栓注入,制成大鼠脑梗死模型;用ELISA方法检测血浆中组织型纤溶酶原激活物(t-PA)及其抑制物(PAI-I)含量;RT-PCR测定大鼠脑组织中t-PAm RNA及其PAI-Im RNA表达的变化。结果与脑梗死对照组[(2.65±0.43)ng/ml]比较,低剂量及高剂量丹参素组t-PA含量[(3.29±0.13)ng/ml、(6.37±0.19)ng/ml]明显升高(均P<0.01),两丹参素组之间差异亦有统计学意义(P<0.01);与脑梗死对照组比较,低剂量丹参素组PAI-I含量[(2.65±0.40)ng/ml]降低(P<0.05),高剂量丹参素组PAI-I含量[(1.98±0.32)ng/ml]明显降低(P<0.01),两丹参素组之间差异亦有统计学意义(P<0.01);与脑梗死对照组比较,低剂量及高剂量丹参素组t-PAmRNA表达[灰度值分别为(0.37±0.14)、(0.46±0.12)]均明显升高(均P<0.01),两丹参素组之间差异亦有统计学意义(P<0.01);与脑梗死对照组比较,低剂量及高剂量丹参素组PAI-ImRNA表达[灰度值分别为(0.54±0.12)、(0.36±0.07)]均明显降低(均P<0.01),两丹参素组之间差异亦有统计学意义(P<0.01)。结论丹参素可以改善脑梗死大鼠纤溶功能,并且有剂量依赖性。 Objective To observe the influence of Danshensu pretreatment on plasminogen activation system in rats with cerebral infarction.Methods 40 male rats were randomly divided into sham operation group,cerebral infarction divided into control group,low-dose Danshensu group [5 mg /(kg·d)] and high-dose Danshensu group [10 mg /(kg·d)],and there were 10 rats in each group.Tissue-type plasminogen activator (t-PA) and its inhibitor (PAI-I) in plasma were detected by enzyme linked immunosorbent assay (ELISA).Expression level of t-PA mRNA and PAI-I mRNA in brain tissue were detected by reverse transcription-polymerase chain reaction (RT-PCR).Results Compared with cerebral infarction control group[(2.65±0.43)ng/ml],the levels of t-PA in low-dose Danshensu group [(3.29±0.13)ng/ml] and high-dose Danshensu group [(6.37±0.19)ng/ml] were increased significantly(all P〈0.01).There was significant difference of the levels of t-PA between the low-dose and high-dose Danshensu groups (P〈0.01).Compared with cerebral infarction control group,the levels of PAI-I in low-dose Danshensu group [(2.65±0.40)ng/ml] was lower (P〈0.05),and in high dose Danshensu group [(1.98±0.32)ng/ml] was significantly lower (P〈0.01).There was significant difference of the levels of PAI-I between the low-dose and high-dose Danshensu groups (P〈0.01).Compared with cerebral infarction control group,the expression levels of t-PA mRNA in low-dose Danshensu group [gray vaule was (0.37±0.14)] and high-dose Danshensu group [gray vaule was (0.46±0.12)] were significantly increased (all P〈0.01).There was significant difference of the levels of t-PA mRNA between the two Danshensu groups (P〈0.01).Compared with cerebral infarction control group,the expression levels of PAI-I mRNA in low-dose Danshensu group [gray vaule was (0.54±0.12)] and high-dose Danshensu group [gray vaule was (0.36±0.07)] were significantly lower (all P〈0.01).There was significant difference of the expression level of PAI-I mRNA between the two Danshensu groups (P〈0.01).Conclusion Danshensu can improve the fibrin dissolve function of the rats with cerebral infarction,and the effect has dose-dependent.
