脑出血大鼠脑组织基质金属蛋白酶-9和S100B表达的变化 被引量:1

摘要 目的研究脑出血大鼠脑组织基质金属蛋白酶-9(MMP-9)和S100B表达的变化。方法将64只SD大鼠随机分为脑出血3d、7d、14d和21d组,每组16只。用自体血注入法制作脑出血大鼠模型,采用免疫组化染色和原位杂交染色的方法,检测各组大鼠脑组织MMP-9、S100B蛋白和mRNA的表达水平。结果脑出血3d组大鼠脑组织MMP-9表达水平明显高于其他3组(均P<0.05),脑出血7d组、14d组和21d组之间差异无统计学意义。脑出血3d组脑组织S100B有少量表达,7d组表达水平与其他3组比较差异有统计学意义(均P<0.05),14d、21d组明显下降。结论 MMP-9和S100B参与了脑出血后脑组织损害的过程,MMP-9是脑出血早期、S100B是脑出血中期脑组织损伤的指标。 Objective To study the changes of expression of matrix metalloproteinase-9 (MMP-9) and S100B of brain tissue in intracerebral hemorrhage(ICH) rats.Methods 64 SD rats were randomly divided into ICH 3 d,7 d,14 d and 21 d groups,and there were 16 rats in each group.ICH models were made by autoblood injection method.The concentration of MMP-9 and S100B including protein and mRNA were detected by immunohistochemical method and in situ hybridization method.Results MMP-9 expression was significantly higher in the ICH 3 d group than that in the other three groups(all P〈0.05).There was no significant difference among ICH 7 d,14 d and 21 d groups.S100B expression was a little in ICH 3 d group,and there were statistical differences between ICH 7 d group and other three groups (all P〈0.05).The expression of S100B were obviously decreased in ICH 14 d and 21 d groups.Conclusions MMPP-9 and S100B are both participate in the pathophysiological processes of brain tissue injury after ICH,MMP-9 is an early stage of ICH and S100B is a intermediate stage of ICH damage criterion of brain tissue.
