人羊水标本处理方法对1H-NMR代谢组学结果的影响

Human Amniotic Fluid Influenced by Different Sampling Methods during ~1H NMR-based Metabonomics Analysis

摘要 对基于氢谱的核磁共振(1H-NMR)的代谢组学方法用于羊水分析进行探讨研究,寻找适合于羊水的试验方法,并建立了一个正常羊水的代谢图谱库,以便下步将羊水用于疾病诊断。设立冻干、冻融、-20℃保存、室温处理四个实验组与对照组进行对比试验,检测其核磁共振图谱,用主成分分析法进行数据分析。结果表明:冻干、冻融次数过多和室温保存过久都会造成羊水中代谢物的变化,影响生物标志物的寻找,-20℃保存一个月和-80℃保存差别不大。所以,基于1H-NMR的代谢组学方法可以用于羊水分析,试验时尽量减少对羊水的处理。 To assess the influence of different sampling methods on Human Amniotic Fluid(HAF) during metabonomics analysis,and to establish a metabolite profile database for normal human amniotic fluid,four experimental groups(the group of freeze-drying,of freeze-thawing,of storage at-20℃,and of keeping in room temperature) and a control group were investigated by use of 1H-NMR spectroscopy,respectively;the data of 1H-NMR spectroscopy was treated by principal components analysis(PCA).The results showed that,by comparison with the control, there were distinet differences in the experimental groups except the group of storage at-20℃.Therefore,It is possible to use 1H-NMR-based metabonomics technique for analysis of HAF;moreover,during the tests,careful treatments of HAF should be institued to minimize the influence on the samples.
