摘要
目的研究白芍总苷(TGP)对成骨及破骨细胞的影响。方法分别取1d龄SD大鼠颅骨及5d龄SD大鼠四肢股骨、胫骨分离培养成骨细胞和破骨细胞。实验分为正常对照组及TGP50,5,0.5,0.05和0.005g.L-1组,采用MTT法检测成骨细胞、破骨细胞活力,金氏法测定成骨细胞培养上清中碱性磷酸酶(AKP)含量。结果 TGP50g.L-1对成骨细胞的存活能力有明显的抑制作用,但对AKP活性影响不明显;TGP5和0.5g.L-1对成骨细胞的存活能力有明显增强作用,使AKP活性明显增强;TGP0.05g.L-1对成骨细胞的存活能力影响不明显,但能明显增强AKP活性;TGP5和0.5g.L-1对破骨细胞存活均有明显的抑制作用。结论白芍总苷可促进成骨细胞存活、增强成骨细胞的AKP活性,过高浓度时则可抑制成骨细胞存活,但对其活性影响不明显;TGP还具有抑制破骨细胞存活的作用。
Objective To observe the effect of total glucosides of peony (TGP) on the proliferation of osteoblasts (OB) and osteoclasts(OC) , the activity of AKP of OB. Methods OB were separated from the skull of SD rats aged 1 d and OC from the thighbone and shinbone of SD rats aged 5 d. The survival of OB and OC was detected by MTT method, and the activity of AKP of OB was examined by King-Amstrong method. Results The proliferation capability of OB was inhibited remarkably by the TGP 50 g·L-1 which could not reinforce the activity of AKP obviously. At the same time, the proliferation capability of OB was strengthened evidently by the 5 g·L-1 and 0.5 g· L-1 which could enhance the activity of AKP clearly. And 0.05 g · L-1 has little influence on the proliferation capability of OB but it can enhance the activity of AKP distinctly. Additionally, the proliferation of OC was inhibited markedly by the TGP 5 g · L-1 and 0.5 g · L-1. Conclusion The TGP could enhance the proliferation of OB and the activity of AKP, but the proliferation capability of OB was inhibited by the hyper concentration TGP, which could not reinforce the activity obviously. The TGP could restrain the proliferation of OC simultaneously.
