摘要
目的观察神经生长因子受体p75 NGFR在分化程度不同的膀胱癌细胞中的表达。方法分别采用免疫组化法,RT-PCR和免疫荧光标记流式细胞术检测法检测p75 NGFR在膀胱癌细胞系SCaBER、T24和5637中的表达。结果免疫组化染色显示p75NGFR蛋白在SCaBER细胞深褐色分布于细胞浆内,而在细胞系T24和5637细胞浆内分别呈褐色和浅褐色分布。p75 NGFR mRNASCaBER、T24、5637中的光密度值分别为(1.40±0.06)、(0.80±0.07)、(0.47±0.09),差异有统计学意义(P<0.05)。SCaBER细胞中p75 NGFR表达阳性者占(9.59±0.25)%,T24细胞占(6.58±0.24)%,5637细胞占(3.11±0.12)%,差异有统计学意义(P<0.05)。结论 p75 NGFR在不同分化程度的膀胱癌细胞中均有表达,表达强弱与肿瘤细胞的分化程度有关。
Objective To investigate the correlation of p75 NGFR expressions and tumor cell differentiation of bladder carcinoma.Methods Immunohistochemical staining,RT-PCR and flow-cytometric measurement were performed to detect the expression levels of p75 NGFR on three cell lines SCaBER,T24 and 5637.Results The expressions of p75 NGFR protein were shown in cytoplasm of three bladder carcinoma cell lines,and p75 NGFR expressions in cell lines SCaBER and T24 was higher than that in cell line 5637.The OD values of mRNA expression levels of p75 NGFR in cell lines SCaBER,T24 and 5637 were(1.40±0.06),(0.80±0.07) and(0.47±0.09) respectively;and the difference among them was significant(P〈0.05).The expression rate of p75 NGFR protein was(9.59±0.25)% in cell line SCaBER,(6.58±0.24)% in cell line T24 and(3.11±0.12)% in cell line 5637,and the difference among them was significant(P〈0.05).Conclusion p75 NGFR is expressed in different tumor cell differentiation of bladder carcinoma,and there is the relationship between p75 NGFR expressions and the degree of malignancy of bladder carcinoma.
