摘要
目的探讨生长抑素受体2(SSTR2)、脆性组氨酸三联体(FHIT)及K-ras蛋白在结直肠癌组织中的表达及相关性,探讨三者与结直肠癌预后的关系。方法应用免疫组化法分别检测18例正常结直肠黏膜、47例溃疡性结肠炎、40例结肠腺瘤及85例结直肠癌组织中SSTR2、FHIT、和K-ras蛋白的表达,分析三者的相关性及与肿瘤临床病理特征的关系。结果 SSTR2、FHIT蛋白在结直肠癌组织中的表达较结肠腺瘤、溃疡性结肠炎、正常结肠黏膜组织低,差异有统计学意义(P<0.05?;K-ras蛋白在结直肠癌组织中的表达较结肠腺瘤、溃疡性结肠炎、正常结肠黏膜组织高,差异有统计学意义(P<0.05?。SSTR2、K-ras蛋白的表达与结直肠癌的分化程度有关(P<0.05),FHIT蛋白的表达与结直肠癌的分化程度、病理分级、Dukes分期有关(P<0.05)。K-ras蛋白的表达与SSTR2和FHIT蛋白的表达呈负相关(r=-0.2198,-0.1720;P<0.05),SSTR2蛋白的表达与FHIT蛋白的表达呈正相关(r=0.3711,P<0.05)。结论 SSTR2、FHIT及K-ras蛋白在结直肠癌组织中有一定程度表达;K-ras蛋白表达升高和SSTR2、FHIT蛋白表达降低与结直肠黏膜癌变有关。
Objective To analyze the correlations among the expressions of somatostatin receptor 2(SSTR2),fragile histidine triad(FHIT),K-ras protein in colorectal cancer tissues;and to investigate their dependability of colorectal cancer prognosis.Methods Using immunohistochemical method,the expressions of SSTR2,FHIT,K-ras proteins in the 18 normal mucous membrane,47 ulcerative colitis,40 colonic adenoma and 85 colorectal cancer tissues were assessed.The correlations between the expressions of SSTR2,FHIT and K-ras proteins;and their relationships to the clinicopathologic characteristics of colorectal cancer were analyzed.Results The expressions of SSTR2 and FHIT proteins in the colorectal cancer tissues were significantly lower than those in normal mucous membrane,ulcerative colitis and colonic adenoma tissues(P〈0.05).The expressions of K-ras protein in the colorectal cancer tissues were significantly higher than those in the normal mucous membrane,ulcerative colitis and colonic adenoma tissues(P〈0.05).The expressions of SSTR2 and K-ras proteins were related with cancer cell differentiation(P〈0.05).The expression of FHIT protein was related with cancer cell differentiation,pathological grade,Dukes stage(P〈0.05).The expression of K-ras protein had negative correlation with the expressions of SSTR2 and FHIT proteins(r=-0.2198,-0.1720;P〈0.05).The expression of SSTR2 protein had positive correlation with the expression of FHIT protein(r=0.3711,P〈0.05).Conclusion SSTR2,FHIT and K-ras proteins are partly expressed in the colorectal cancer tissues;the up-regulation of K-ras protein and the down-regulations of SSTR2 and FHIT proteins are related with the cancerization of colonic mucous membrane.
