摘要
为研制新型抗肿瘤药物,以3,4-二甲氧基苯乙腈和对甲氧基苯甲醛合成白藜芦醇类似物,并以MTT法、流式细胞仪细胞周期分析、Hoechst33258荧光染色及Western blot方法检测合成的白藜芦醇类似物对肝癌Bel-7404细胞的增殖影响及其机制。实验结果证明,所合成的含甲氧基白藜芦醇类似物可通过诱导Bcl-2蛋白表达下调、Bax蛋白表达上调,引起Bel-7404细胞G2/M周期阻滞,并发生细胞凋亡,最终抑制细胞增殖。
In order to find a new anticancer-drug,a resveratrol analogue is synthesized with 3,4-dimethoxyphenyl-acetonitril and p-methoxybenzaldehyde,and the hepatocellular carcinoma Bel-7404 cells are treated with the resveratrol analogue.The treated sample are detected by MTT,Flow Cytometer,Hoechst 33258 Fluorescence Coloration and Western blot,in order to examine the influence of the analogue on the proliferation of the Bel-7404 cells and its mechanism.The results show the resveratrol analogue with methoxy can induce the decrease in the expression of Bcl-2 and the increase in the expression of Bax,stop cell cycle in G2/M,make the apoptosis happen and finally inhibit the proliferation of Bel-7404 cells.
