摘要
目的探讨缝隙连接蛋白43(Cx43)在前列腺肿瘤中的生物学意义。方法免疫组化法观察Cx43在正常组织和肿瘤组织以及体外培养的前列腺癌PC-3细胞株中的表达,并采用半定量RT-PCR法检测mRNA水平的变化。结果本实验在mRNA水平上证实PC-3细胞有Cx43 mRNA的表达,在蛋白水平上证实PC-3细胞和前列腺癌组织均有Cx43的表达,但其表达较正常者减弱,蛋白异常定位于胞浆中而非胞膜上。而且Cx43在前列腺癌组织中的表达与前列腺癌病理分级呈负相关。结论Cx43降低可能影响前列腺癌组织及前列腺癌细胞的发生和发展。
Objective To investigate the expression of Cx43 in Prostate cancer and its biological signicicance.Methods The expression of Cx43 was detected by immunohistochemistry and semi-quantity-RT-PCR in 15 normal prostate tissues,35 prostate cancer tissues and the PC-3 cell line.Results The expression of Cx43 protein was intensive in normal prostate tissues,and it was clearly located on the plasma membrane,but faint or negative in 74.2% of cancer tissues.The expression of Cx43 protein was reduced in early highly differentiated prostate cancers and PC-3 cells,and was negative to the malignant type.In addition,the expression of Cx43 mRNA was also decreased in tumors(P0.05).Conclusion The expression of Cx43 is evidently decreased in prostate cancers,which may induce the reduced function of GJIC.
