摘要
目的研究水通道蛋白-4(AQP_4)和血管内皮生长因子(VEGF)在视网膜缺血/再灌注(RIR)损伤机制中的表达规律,探讨二者在RIR损伤机制中有无相关性。方法运用提高眼内压的方法建立大鼠RIR模型,将56只Wistar大鼠随机分为术后6、12、24、48、72 h,7d组和正常对照组,用免疫组化法、平均光密度分析和Pearson法观察二者的表达水平及相关性。结果 AQP_4在RIR损伤6 h后出现表达上调,24 h后达高峰,除7 d组外,其余实验组AQP_4的表达与正常对照组相比均有显著性差异(P<0.01)。VEGF表达在RIR损伤后12 h开始增加,48 h达高峰,具有显著性差异(P<0.01)。AQP_4和VEGF在12~72 h呈正相关,且24、48 h的相关性更佳。结论 AQP_4和VEGF在RIR损伤中可能通过协同作用影响视网膜的水代谢。
ObjectiVe To investigate the regular pattern of expression and to analyze the relationship between aquaporin-4 (AQP4) and vascular endothelial growth factor (VEGF) in the retinal ischemia/reperfusion (RIR) injury. Methods The model of transient ischemia/reperfusion injury of the rat retina was constructed by elevation of the intraocular pressure. Fifty six Wistar rats were randomly divided into 6, 12, 24, 48, 72 h and 7 d group after retinal ischemia and normal control group, with immunohistochemical staining and average optical density analysis and to observe the expression level and their correlation. There was significant correlation between AQP4 and VEGF from 12 h to 3 d, and the correlations of 24 h and 48 h were better than others. Results AQP4 began to express after 6 h in ischemia group, increased gradually and reached maximal level in 24 h after RIR injury, except in 7 d group the difference was significant (P 〈0.01). VEGF began to express after 12 h in ischemia group, and reached maximal level in 48 h after RIR injury and the difference was significant (P 〈0.01). Conclusion AQP4 and VEGF may co-affect the water metabolism of retina by cooperation in the RIR injury.
