摘要
目的:探讨单纯疱疹病毒I型(HSV-1)感染后在宿主骨髓是否形成潜伏感染。方法:尾静脉注射HSV-1感染小鼠,分别于病毒接种后5、15、30 d处死实验组和对照组小鼠,取股骨分离培养原代骨髓间充质干细胞(BMSCs),并对其进行成骨细胞诱导和树突状细胞诱导的鉴定,PCR法检测骨髓及BMSCs中HSV-1特异条带。结果:BMSCs成骨诱导碱性磷酸酶和钙结节染色均呈阳性,培养出形态典型的树突状细胞。各实验组骨髓及其对应的BMSCs的第1、2代均检测到HSV-1的特异DNA片段。结论:骨髓是HSV-1形成潜伏感染的靶器官之一,且能随细胞增殖而持续携带HSV-1。
Objective: To study whether herpes simplex virus type 1 (HSV-1) could cause latent infection on the host bone marrow. Methods: Mice of the experimental group and control group were sacrificed respectively in the 5th day,the 15^th day and the 3^th day after tail vein injection of HSV-1, and the primary bone marrow-derived mesenchymal stem ceils (BMSCs) were cultured and identified. The specific gene fragments of HSV-1 were examined by PCR in bone marrow and BMSCs. Results: The cells obtained from BMSCs were identified to be osteoblasts by alkaline phosphatase stain and alizarin bordeaux stain of calcified nodule; The typical morphology of dendritic cells which also obtained from BMSCs were cultured. The specific gene fragments of the experimental group and its corresponding first and second generation BMSCs were amplified correctly by PCR. Conclusion: Bone marrow is one of the organs that could be infected by HSV-1 which could consistently exist with cell proliferation.
