摘要 目的:建立糖尿病牙周炎大鼠模型以利于研究牙周炎与糖尿病的关系。方法:2月龄雄性Wistar大鼠20只随机分成糖尿病牙周炎大鼠组即实验组(n=10)和空白对照组(n=10)。分别给予高糖软食,普食喂养。实验组大鼠大剂量STZ枸橼酸缓冲液腹腔注射,并用正畸钢丝加丝线结扎下颌第一磨牙;对照组不做处理。实验第6周处死两组大鼠,观察大鼠下颌磨牙牙周组织病理学变化。结果:注射STZ枸橼酸缓冲液3 d后实验组大鼠出现多饮、多食、多尿、体重下降的症状。用正畸结扎丝加丝线牙周结扎后实验组出现食物嵌塞、牙龈探诊出血、唾液较多、牙周袋形成、牙槽骨吸收等症状,与牙周炎相符。对照组大鼠则无上述表现。两组大鼠6周后的血糖值,牙周袋探诊深度值比较有显著差异(P<0.01)。结论:建立了稳定的糖尿病牙周炎模型,为糖尿病与牙周炎关系的进一步研究提供了很好的方法。 Objective: To establish a diabetic periodontitis model for the study of the relationship between periodontitis and diabetes. Methods: 20, two-month-old male Wistar rats were randomly divided into diabetic periodontitis group (experimental group, n=10) and control group (n=10). Rats were given high-sugar soft food, general food respectively feeding.The model was established by intraperitoneal injection of streptozotocin (STZ) and by ligating floss around maxillary frist molars with orthodontic wire ligature wire. Six weeks later, animals were sacrificed and the specimens were prepared for histological analysis. Results: The Polydipsia, polyphagia, polyuria, weight loss were observed three days after injection of STZ citrate buffer in the experimental group rats. Food impaction, saliva more, periodontal pocket formation, alveolar bone loss and other symptoms were also observed. Rats of control group had no periodontitis in the performance of the above. Compared with control group rats in experimental group had more higher hyperglycemia and more periodontal pocket formation (P〈0.01). Conclusion: The method can establish a stable model of diabetic periodontitis. It is provided a good way in the further study between diabetes and periodontitis.
