二甲双胍对多囊卵巢综合征大鼠卵巢肿瘤坏死因子-α表达及血清激素的影响 被引量:4

摘要 目的观察二甲双胍对多囊卵巢综合征(PCOS)大鼠卵巢组织肿瘤坏死因子-α(TNF-α)表达和血清激素的影响,探讨其对PCOS大鼠的治疗机制。方法应用来曲唑灌胃建立PCOS大鼠模型,选取6周龄清洁级雌性Wistar大鼠30只,随机分为对照组、PCOS组、二甲双胍治疗组,每组10只。采用放射免疫法测定血清性激素水平,光镜观察卵巢、卵泡发育情况,血糖仪测定糖耐量,免疫组织化学法测定卵巢组织TNF-α的表达水平。结果二甲双胍可使PCOS大鼠卵巢颗粒细胞层增厚,卵泡膜细胞层变薄,卵巢组织TNF-α的表达明显减弱,血清睾酮(T)、黄体生成素(LH)、空腹血糖(FPG)及空腹胰岛素(FIN)水平降低。结论二甲双胍能够通过影响卵巢TNF-α的表达调整内分泌,进而调整PCOS大鼠性腺功能,促进卵泡的发育成熟,恢复正常排卵。 Objective To observe the effects of metformin on expression of ovarian tumor necrosis factor-alpha (TNF-α) and serum hormone in rats with polycystic ovarian syndrome (PCOS), and to investigate its therapeutic mechanism in PCOS rats. Methods Letrozole gastric perfusion was used for establishing PCOS rat model. Thirty female Wistar rats at 6 weeks of life were randomly divided into control group, PCOS group and metformin group (n=10 in each group). Levels of serum sex hormone were detected by radioimmunoassay, and ovaries and ovarian follicular development were observed by light microscope, with oral glucose tolerance test (OGTT) performed. Moreover, immuno- histochemistry was used to detect the expressed level of TNF-α. Results Mefformin thickened ovarian granular cell layer in PCOS rat, and thinned theca cell layer. Besides, it greatly reduced the expression of TNF-α, and also decreased the levels of testosterone (T), luteinizing hormone (LH), fasting plasma glucose (FPG) and fasting insulin (FIN). Conclusion Mefformin was shown to regulate expression by modulating expression of ovarian TNF-α, thereby improving gonadal functions, promoting maturation of follicles and recovering normal ovulation in PCOS rats.
