摘要
目的:探讨牙齿正常状态和炎症过程早、中、晚期胶质源性神经营养因子(glialcelline-derivedneurotrophicfactor,GDNF)的基因表达变化。方法:在大鼠牙髓炎模型上,分别摘提取正常鼠及炎症鼠早、中、晚期的尾侧亚核,提取总RNA,用RT-PCR一步法扩增GDNFmRNA,琼脂糖凝胶电泳照像。结果:正常鼠尾侧亚核GDNFmRNA有一定水平的基础量表达;炎症急性期GDNFmRNA表达量略有下降;炎症修复期GDNFmRNA表达量上升至高峰后回落至高于基础表达量的水平。结论:牙髓炎修复期GDNF在尾侧亚核F的表达增高,提示GDNF可能参与炎症牙髓神经修复再生过程,对神经末稍再生发芽有促生长作用。
Aim: Investigation of the expression of GDNF mRNA of caudat subnucleus in different stages of rat inflammation dental pulp. Methods: Model of rat inflammation dental pulp was established and total RNA was extracted from caudat subnucleus in different stages of rat inflammation tooth. The desired DNA product was obtained from total RNA by RT-PCR. cDNA was amplified and showed on 2% agarose. Results: A 337 bp band was showed on 2% agarose. The extension of light was strong in middle stages of rat inflammation dental pulp,while it was weak in earlier stage and later stage. Conclusion: The results suggest that GDNF may take part in regulating nerve-sprouting reactions during dental restorative process.
