肿瘤细胞与正常细胞共有大量的强抗原,而大多数肿瘤相关抗原则相对较弱,这就给用肿瘤细胞免疫BALB/c小鼠以研制抗肿瘤单克隆抗体(McAb)的工作带来了不少困难。Watanabe等用免疫功能重建的方法获得了抗胃癌的McAb。

Spleen cells of nude mice bearing human lung adenocarcinoma were fused with mouse m eloma cell ling NSO. Hybridoma cells gre in 98% of wells of eight cell culture plates. Four monoclonal antibodies, ALA-01, 02, 03 and 04, very strongl reacted with human lung adenoeareinoma cell lines, LAX and Lu. apa, but did not cross-react with various human normal tissues and the other kinds of tumor cell lines using ELISA and immunofluorescence stain. These hybridoma cells were able to induce ascitcs after inoculating the cells into BALB/c mice. Immunodiffusion assay indicated that all of them were IgG_1 types. Immunofluorescence and immunohistochemistry stain against four samples from patienis with lung adenocarcinoma shoedw that ALA-01 could specially bind membrane of the cancer cells.
