摘要
目的:探讨通过体内阻断c-kit信号途径来建立豚鼠膀胱Cajal样细胞缺失模型的方法。方法:选取新生豚鼠20只,随机分成实验组、对照组。实验组在出生24h内的豚鼠腹腔内隔天注射c-kit抗体(ACK2)100μg,共5次,对照组腹腔内注射生理盐水,于第10天取膀胱。然后将两组豚鼠膀胱制作冰冻切片行免疫荧光染色及激光共聚焦显微镜观察。结果:10d后实验组与对照组豚鼠逼尿肌中Cajal样细胞在激光共聚焦显微镜下比较:实验组膀胱逼尿肌中Cajal样细胞数量减少甚至消失(P<0.01),Cajal样细胞二聚体消失(P<0.01)。结论:通过阻断c-kit信号通路可以成功建立豚鼠膀胱Cajal样细胞缺失模型。
Objective:To establish a new model of interstitial cell of Cajal-like cells loss in the guinea-pig bladder.Methods:New-born guinea-pigs were injected intraperitoneally antibody of C-kit(ACK2) 100μg every second day,and control groups were injected saline.The distribution of Cajal-like cells was observed by laser scanning confocal microscope when the guinea-pigs were 10 days postpartum.Results:The distribution of interstitial cell of Cajal-like cells was lost when C-kit was blocked.Conclusion:When the C-kit signal is blocked,interstitial cell of cajal-like cells loss is found in the guinea-pig bladder.
