镇江农村饮用水有机提取物致突变性研究 被引量:2

摘要 目的了解镇江肿瘤高发村生活用水污染物致突变强度,为判断肿瘤病因提供依据。方法选取3个肿瘤高发村为观察区,3个环境相似的村为对照区。应用鼠伤寒沙门菌组氨酸回复突变试验(Ames)试验,测试生活饮用水提取物对鼠伤寒沙门菌的组氨酸营养缺陷型菌株回复突变强度。结果对照区3个村和观察区的前隍村水样有机提取物在加与不加S9时所设剂量有机提取物诱导的回复突变菌落数都没有超过自发对照的2倍,结果为阴性。观察区的江心洲1号、2号河水有机提取物在加S9与不加S9时对TA98、TA97试验菌株诱发回复突变的MR值分别为2·71、3·38和2·71、3·00。1号、2号出厂水样有机提取物在加S9时对试验菌株TA98、TA97诱发回复突变的MR值分别为2·18、2·00和2·06、2·00,存在明显剂量-反应关系。研究区的勤丰村1号、2号河水有机提取物在加S9与不加S9时对TA98、TA97、TA100试验菌株诱发回复突变的MR值分别为4·30、4·70,2·32、2·44,2·00、2·36和3·39、3·79,2·18、2·29,2·00、2·14。勤丰村1号、2号河水有机提取物在加S9时对TA102试验菌株诱发回复突变的MR值分别为2·07、2·01。勤丰村1号、2号浅井水有机提取物在加S9时对TA98、TA97、TA100试验菌株诱发回复突变的MR值分别为2·03、2·15、2·02和2·03、2·14、2·00,存在剂量反应关系。结论镇江农村饮用水有机提取物具有致突变性,饮用水有机污染可能是镇江农村肿瘤高发的主要危险因素之一。 Objective To understand the mutation intensity caused by water pollution in the villages with high incidence of cancer in Zhenjiang and provide evidence to identify the cancer cause.Methods The Ames test was conducted to detect the reverse mutation intensity of water extract to histidine auxotrophic strains of Salmonella typhimurium in 3 villages with high incidence of cancer(study area)and in 3 villages with similar environment(control area).Results The mutant colonies with reverse mutation induced by organic extracts in water samples from 3 control villages and Qianhuang village in study area(with S9 or without S9)did not exceed the self-control by 2 times,the results were negative.The MR values of reverse mutation for tested strains(TA98,TA97)induced by the organic extracts from the water in No.1 and No.2 rivers in Jiangxinzhou(with S9 or without S9)were 2.71,3.38 and 2.71,3.00 respectively.The MR values of reverse mutation for tested strains(TA98,TA97)induced by the organic extracts from the water in No.1 and No.2 water factories in Jiangxinzhou(with S9)were 2.18,2.00 and 2.06,2.00 respectively.There was obvious dose-response relationship.The MR values of reverse mutation for tested strains(TA98,TA97 and TA100)induced by the organic extracts from the water in No.1 and No.2 rivers in Qinfeng village(with S9 or without S9)were 4.30,4.70,2.32,2.44,2.00,2.36 and 3.39,3.79,2.18,2.29,2.00,2.14 respectively.The MR values of reverse mutation for tested strain(TA102)induced by the organic extracts from the water in No.1 and No.2 rivers in Qinfeng village(with S9)were 2.07,2.01 respectively.The MR values of reverse mutation for tested strains(TA98,TA97 and TA100)induced by the organic extracts from the water in No.1 and No.2 shallow wells in Qinfeng village(with S9)were 2.03,2.15,2.02 and 2.03,2.14,2.00 respectively.There was obvious dose-response relationship.Conclusion The organic extracts from drinking water in rural area in Zhenjiang showed mutagenicity.The organic pollution of drinking water might be the one of the major risk factors of high incidence of cancer in rural areas in Zhenjiang.
