多肽固相序列分析载体的合成及应用

摘要 用合成的聚合物作载体,与模型15肽(纯品)偶联,使用Edman降解法进行多肽序列分析。比较了各种类型载体对肽的偶联效果。实验结果表明合成的大孔型四次乙基五胺类树脂载体已经达到国外同类产品水平。 Several polymer supports for sequence analysis were derived from low crosslinked gelatinous and macroporous poly(styrene-divinylbenzene). Three types of functionalized supports have been compared with each other for coupling model peptide and subsequent Edman degradation.The experimental results indicated that under suitable functional group content of about 1.0 m mol NH_2/g of resin, the attaching yields of peptide to macroporous resin were higher than that of the gel type of resin. Macroporous TEPA resin has been demonstrated to be qualified as a polymer support for solid-phase sequence analysis of peptide.
