油菜花粉有效成分山奈酚在家兔体内的药代动力学研究 被引量:3

摘要 目的:采用高效液相色谱法研究健康家兔被灌胃油菜花粉(15g/kg)后的山奈酚药代动力学。方法:对动物给药后,在不同时间分点经颈静脉采血,血浆样品的酶解物通过C18小柱富集,对甲醇洗提部分进行色谱分析。药代动力学参数运用PKS软件处理。结果:家兔被灌胃油菜花粉后山奈酚浓度-时间曲线符合一级吸收二室模型。主要药动学参数:AUC(1.4599±0.4389)(μg·h)/L、Tpeak(3.7143±0.9512)h、Cmax(0.2455±0.0615)μg/mL、T1/2β(1.99±0.19)h、Vd(31.418±8.010)L/kg。结论:家兔被灌胃油菜花粉后,山奈酚具有吸收较缓慢,体内分布广,消除迅速的特点。 Objective:An HPLC method was used for the determination of Kaempferol in rabbit plasma following oral administration at a single dose of 15 g/kg to study its pharmacokinetics features.Methods:Blood samples were collected from jugular vein at different time points,plasma samples that were enzymatic hydrolyzed were loaded onto a C18 column,the methanol eluate was analyzed by using HPLC.The pharmaceutical kinetics parameter of Kaempferol were treated with PKS Software.Results:The best pharmacokinetic model of Kaempferol was two-compartment open model.The main pharmacokinetic parameters were as follows:AUC(1.4599±0.4389)(μg·h)/L、Tpeak(3.7143±0.9512)h、Cmax(0.2455± 0.0615)μg/mL、T1/2β(1.99±0.19)h、Vd(31.418±8.010)L/kg,respectively.Conclusion:After the rabbits were orally administrated with rape bee pollen,Kaempferol was slowly absorbed,rapidly eliminated,and possessed large volume of distribution.
