葛根芩连微丸体外抗轮状病毒作用实验研究 被引量:7

摘要 目的:评价葛根芩连微丸体外干预对人轮状病毒A组G3型709株的药理作用。方法:通过细胞病变效应、噻唑蓝比色法检测细胞活性,观察葛根芩连微丸抗人轮状病毒709株的作用。结果:葛根芩连微丸的恒河猴胚胎肾细胞半数存活浓度为0.703mg/mL,对人轮状病毒709株无直接灭活杀伤作用;对人轮状病毒709株有一定预防吸附的作用,IC50=0.146mg/mL,TI=4.815068;能够抑制人轮状病毒709株细胞内生物合成的作用,IC50=0.023mg/mL,TI=30.56522。结论:葛根芩连微丸对轮状病毒吸附宿主细胞有一定干预作用,但是主要通过抑制轮状病毒在恒河猴胚胎肾细胞内的生物合成过程发挥体外抗轮状病毒作用。 Objective:To evaluate Gegenqinlian micropellet’s pharmacological effect of the direct destruction of human rotavirus,human rotavirus invasion of host cells and the inhibition of the biological synthesis of human rotavirus in host cells in vitro.Methods: By improved methods of MTT and CPE of Institute of Virology School of Medicine Wuhan University to evaluate the biological effect of Gegenqinlian micropellet on rotavirus. Results: The half toxic concentration of Gegenqinlian micropellet was 0.703 mg/mL. The 50% inhibitory rotavirus biological synthesis of concentration of Gegenqinlian micropellet was 0.023mg/mL. Conclusions:In vitro Gegenqinlian micropellet can not directly kill human rotavirus,while can not only prevent human rotavirus invasion of host cells,but also can inhibit rotavirus in host cells within the biological synthesis of the process. The latter effect is much more important during the anti-rotavirus procedure.
