摘要
c-myc原癌基因是细胞增殖调控中的一种关键基因。文中报道了硫代磷酸修饰的c-myc反义寡核苷酸(ODNS)对血小板源性生长因子(PDGF)刺激的肺动脉平滑肌细胞(SMC)增殖及c-myc基因表达的影响。Northem杂交结果表明:反义ODNS能显著下调PDGF刺激的c-myc基因表达:3H-TdR试验及细胞生长分析表明:反义ODNs能显著抑制PDGF刺激的SMC增殖。同义00Ns无上述作用。
C-myc oncogene plays an important role in the proliferation of cells.The present studyexamined the effects of c-myc phosphorothioate oligodeoxynucleotides (ODNs) on c-myc expressionand proliferation of pulmonary arterial smooth muscle cells (SMC) stimulated by PDGF. Northernblot analysis showed that antisense-ODNs reduced c-myc expressions significantly.3 ̄H-TdR incor-poration and cell proliferation assay showed that antisense-ODNs significantly inhibited proliferation ofSMC.Control sense-ODNs did not affect c-myc expression and proliferition of pulmonary arterialSMC.
