Effect of c-myc Antisense Oligodeoxynucleotide on Proliferation ofPulmonary Arterial Smooth Muscle Cells Stimulated by PDGF

ｃ－ｍｙｃ原癌基因是细胞增殖调控中的一种关键基因。文中报道了硫代磷酸修饰的ｃ－ｍｙｃ反义寡核苷酸（ＯＤＮＳ）对血小板源性生长因子（ＰＤＧＦ）刺激的肺动脉平滑肌细胞（ＳＭＣ）增殖及ｃ－ｍｙｃ基因表达的影响。Ｎｏｒｔｈｅｍ杂交结果表明：反义ＯＤＮＳ能显著下调ＰＤＧＦ刺激的ｃ－ｍｙｃ基因表达：３Ｈ－ＴｄＲ试验及细胞生长分析表明：反义ＯＤＮｓ能显著抑制ＰＤＧＦ刺激的ＳＭＣ增殖。同义００Ｎｓ无上述作用。

C-myc oncogene plays an important role in the proliferation of cells.The present studyexamined the effects of c-myc phosphorothioate oligodeoxynucleotides (ODNs) on c-myc expressionand proliferation of pulmonary arterial smooth muscle cells (SMC) stimulated by PDGF. Northernblot analysis showed that antisense-ODNs reduced c-myc expressions significantly.3￣H-TdR incor-poration and cell proliferation assay showed that antisense-ODNs significantly inhibited proliferation ofSMC.Control sense-ODNs did not affect c-myc expression and proliferition of pulmonary arterialSMC.
