摘要
目的 观察人脑胶质瘤组织的低密度脂蛋白受体(LDLR)活性和其mRNA水平,探讨其与胶质瘤肿瘤级别的关系.方法 以125I-LDL为配体,采用受体-放射性配体结合分析法测定63例肿瘤及其周边脑组织的LDLR活性;逆转录-聚合酶链反应(RT-PCR)技术检测LDLR蛋白mRNA在瘤组织的表达.结果 胶质瘤组织LDLR活性为(129.43±48.04)μmol/g组织,其周边正常脑组织LDLR为(34.06±27.62)μmol/g组织,两者比较差异有统计学意义(P<0.01);高级别胶质瘤组织的LDLR为(154.59±31.68)μmol/g组织,低级别胶质瘤组织LDLR活性为(92.32±24.37)μmol/g组织,高级别胶质瘤LDLR活性高于低级别(P<0.05);胶质瘤组织LDLR蛋白mRNA水平均增高,但组间差异无统计学意义(P>0.05).结论 脑胶质瘤组织LDLR活性明显增高,肿瘤级别越高,LDLR活性越高;因此,LDLR有可能成为脑胶质瘤治疗的潜在靶点.
Objective To observe the activity of low-density lipoprotein receptor (LDLR) and the expression of LDLR mRNA in glioma, as well as to analyze the relationship between the activity of LDLR and the tumor grade of glioma. Methods Using 125I-LDL as the radioligand, method of receptor-radioligand binding analysis was used to determine the LDLR activity of 63 patients with glioma in glioma tissue and brain tissue. The expression of LDLR mRNA in glioma and brain tissue was examined by reverse transcription-polymerase chain reaction (RT-PCR). Results The uptake of 125I-LDL was ( 129.43 ±48.04)μmol/g tissue in glioma tissue and (34.06 ± 27. 62) μmol/g in the normal brain tissue ( P 〈 0. 01 ). The prevalence of 125I-LDL uptake was ( 154. 59 ± 31.68 ) μmol/g tissue in high-grade glioma and ( 92. 32 ±24. 37 ) μ mol/g tissue in low-grade tumor ( P 〈 0. 05 ). The expression of LDLR mRNA was significantly increased in glioma, but there was no significant difference in the expression of LDLR mRNA in the tissue of glioma between low-and high-grade tumors ( P 〉 0.05 ). Conclusion The activity of LDLR in glioma was obviously higher, and the higher the tumor grade of glioma, the higher the activity of LDLR. The expression of LDLR mRNA had no significant difference between low- and high-grade gliomas.
