负载表柔比星的磁性壳聚糖纳米粒CS-PAA-EPI杀伤人膀胱癌T24细胞效应及其机制

摘要 目的 探讨负载表柔比星的磁性壳聚糖纳米粒CS-PAA-EPI对人膀胱癌T24细胞的杀伤效应及机制.方法 选用壳聚糖、磁流体及表柔比星为原材料首次合成载表柔比星的磁性壳聚糖纳米粒,运用噻唑蓝(MTT)比色法、流式细胞术等技术检测CS-PAA-EPI磁性纳米粒对人膀胱癌T24细胞的杀伤效应,并通过凋亡相关蛋白的检测对其机制进行初步探讨.结果 实验组对T24细胞的生长抑制率分别为76.6%、71.8%、68.4%,均明显高于对照组53.8%(P<O.05),实验组细胞凋亡率为55.13%,并且在亚二倍体区(sub-G1)出现明显"凋亡峰".结论 CS-PAA-EPI磁性纳米粒可以有效杀伤人膀胱癌T24细胞,其杀伤机制可能是通过诱导细胞凋亡引起. Objective To investigate the effects and mechanism of CS-PAA-EPI polymer magnetic nanopaticle on human bladder cancer T24 cells. Methods Magnetic CS-PAA-EPI Nanopaticle was composed by CS,acrylic monomer (AA), EPI and Fe3O4 nanopaticle. The growth inhibiting rate of T24 cells were detected by the MTT techniche, apoptosis of T24 cells were observed by flow cytometry in vitro. The protein expression of T24 cells were tested by Western blotting techniche. Results To compare with control group, the growth inhibiting rate (76.6%, 71.8%, 68.4%) and apotosis rate (55. 13% ) of T24 cells in experimental group were significantly increased ," apoptosis peak" was observed in sub-G1 of experimental group cells. Conclusion CS-PAA-EPI Polymer Magnetic Nanopaticle can effectively kill human bladder T24 cells, the potential mechanism is cell apotosis.
