摘要
为了快速测定真菌油脂的含量,对油脂含量的测定方法进行了研究。建立了一种真菌油脂含量快速测定方法,这种方法是将湿菌体经微波预处理后,直接皂化,生成甘油,然后再将甘油直接转化为三乙酸甘油酯,用气相色谱法分析生成的三乙酸甘油酯以检测菌体中油脂的含量。试验结果表明,所建立的真菌油脂含量快速测定方法的精密度及可靠性良好,该方法的回收率范围为97.99%~104.96%,变异系数不超过3.76%。该方法中,微波预处理的功率为420W,时间为2min;微波强化皂化的功率为140w,时间为4min;甘油酯化时间为10min。
Rapid detection method of the oil content in fungi was studied. The wet mycelium pretreated by microwave with the power of 420 W and the time of 2 min was treated directly with KOH -CHaOH under the microwave treating conditions of the power of 140 W and the time of 4 min to saponify the oil, and the glycerol obtained from the saponification was esterified for 10 min to obtain glycerol triacetate, then GC was employed to identify the glycerol triacetate so as to determine the oil content in mycelium. The results showed that the coefficient of recovery of the rapid detection method was 97.99% - 104.96% , and the coefficient of variation was less than 3.76%. The method of rapid detection possessed the characters of a good precision, accuracy and reliabihty.
