产超广谱β-内酰胺酶大肠埃希菌质粒谱与基因型的相关性

Relation between plasmid map and extended spectrum β-lactamases genotype of producing ESBLs in E. coli

目的:探讨产超广谱β-内酰胺酶(ESBLs)大肠埃希菌质粒谱与ESBLs基因型的相关性。方法:收集郑州大学第一附属医院检验科2007年至2008年临床分离产ESBLs大肠埃希菌46株,采用琼脂糖凝胶电泳进行质粒谱分析,采用PCR技术检测ESBL基因型,分析同一菌株所携带质粒条带数与其携带ESBLs基因型别数的相关性。结果:产ESBLs大肠埃希菌有5种质粒谱型(分别为携带单一21000bp质粒、21000bp+2500bp2种质粒、21000bp+2500bp+3000bp3种质粒、21000bp+2500bp+3000bp+7000bp4种质粒以及21000bp+2500bp+3000bp+7000bp+9000bp5种质粒)和5种ESBLs基因型(1种型别10株,2种型别15株,3种型别12株,4种型别8株,5种型别1株),同一菌株质粒谱与其ESBLs基因型无相关性(rs=0.091,P=0.548)。结论:多种ESBLs表达基因可定位于同一质粒上。

Aim： To discuss the relation between the plasmid map and extended spectrum β-lactamases（ESBLs） genotype of produeing ESBLs E. coli. Methods：A total of 46 strains of producing ESBLs E. coli collected from the first affiliated hospital of Zhengzhou university during 2007 to 2008. Plasmid maps were detected by agarose electrophoresis. Results：5 plasmid maps（contained 21 000 bp,21 000 bp ＋2 500 bp,21 000 bp ＋2 500 bp ＋3 000 bp,21 000 bp ＋2 500 bp ＋3 000 bp＋7 000 bp,21 000 bp＋2 500 bp＋3 000 bp＋7 000 bp＋9000 bp,respectively） and5 ESBLs genotypes （1 genotype 10 strains,2 genotypes 15 strains,3 genotypes 12 strains,4 genotypes 8 strains,5 genotypes 1 strains） were found. The plasmid maps have no correlation to ESBLs genotype in 1 strains（ r = -0. 091,P = 0. 548）. Conclusion： All kinds of ESBLs expressing genes may locate at one plasmid.
