摘要 目的:观察三磷酸腺苷结合盒转运体8(ABCG8)T400K基因多态性与辛伐他汀降脂效果的关系。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法对以辛伐他汀治疗的182例高血脂患者进行基因分型,测定治疗前后血清甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL-C)及高密度脂蛋白(HDL-C)浓度的变化。结果:TT型携带者138例,TK型携带者41例,KK型携带者3例,ABCG8400K等位基因分布频率为12.91%。辛伐他汀治疗前后K等位基因携带组血脂浓度的变化率与TT型携带组相比差异无统计学意义[TG:(24.09±10.30)mol/Lvs(26.13±10.82)mol/L,t=1.098,P=0.274;TC:(18.13±3.78)mol/Lvs(18.48±3.85)mol/L,t=0.532,P=0.595;LDL-C:(27.34±6.34)mol/Lvs(27.39±5.69)mol/L,t=0.046,P=0.963;HDL-C:(0.45±9.08)mol/Lvs(2.48±12.86)mol/L,t=0.973,P=0.332]。结论:ABCG8T400K基因多态性与辛伐他汀降脂效果无关。 Aim:To investigate the association between ATP binding cassette transporter 8(ABCG8)T400K polymorphism and lipid-lowering response to simvastatin.Methods:Genotypes were determined using PCR-RFLP in 182 hyperlipidemic patients treated with simvastatin 20 mg once daily for 30 days.Serum triglyceride (TG),total cholesterol (TC),low-density lipoprotein cholesterol (LDL-C),and high density lipoprotein cholesterol (HDL-C) levels were determined before and after treatment.Results:Among the 182 genoeyped subjects,138 were TT carriers,41 TK carriers,and 3 KK carriers.The frequency of the ABCG8 T 400K variant allele was 12.91%.After Simvastatin treatment,the difference in the percentage change of serum lipid levels was not statistically significant between carriers of the T400K variant and noncarriers,[TG:(24.09±10.30) mol/L vs (26.13±10.82) mol/L,t=1.098,P=0.274;TC:(18.13±3.78) mol/L vs (18.48±3.85) mol/L,t=0.532,P=0.595;LDL:(27.34±6.34) mol/L vs (27.39±5.69) mol/L,t=0.046,P=0.963;HDL:(0.45±9.08) mol/L vs (2.48±12.86) mol/L,t=0.973,P=0.332].Conclusion:There is no association between ABCG8 T400K polymorphism and lipid-lowering response to simvastatin.
