摘要
目的:检测儿童先天性巨输尿管组织中缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)和转化生长因子-β1(TGF-β1)蛋白的表达。方法:取25例先天性巨输尿管末端组织,按年龄分为<1岁组(10例),1~3岁组(8例)和>3岁组(7例);取28例肾母细胞瘤输尿管下端无肿瘤细胞浸润的组织作为对照,按年龄分为<1岁组(8例),1~3岁组(10例)和>3岁组(10例)。采用免疫组化SP法检测两种组织中HIF-1α、VEGF和TGF-β1蛋白的表达。结果:>3岁组对照组织和先天性巨输尿管末端组织中HIF-1α、VEGF和TGF-β1的表达极少或呈阴性。<1岁组和1~3岁组先天性巨输尿管末端组织中3指标均高于对照组织(HIF-1α:Z=3.593和3.721;VEGF:Z=3.554和3.465;TGF-β1:Z=3.554和3.552,P均<0.001)。结论:HIF-1α、VEGF和TGF-β1的异常表达可能参与了儿童先天性巨输尿管症的发病过程。
Aim:To study the expressions of HIF-1α,VEGF and TGF-β1 in primary megaureter tissue of children.Methods:A total of 25 specimens of megaureter ending were obtained from children and divided into 3 groups:1 year old group(n=10),1~3 years old group(n=8) and 3 years old group(n=7).Control group:inferior segment of ureter specimens were taken from 28 cases of renal neoplasm which were confirmed histologically to be unaffected,and they were also divided into 3 groups:1 year old group(n=8),1~3 years old group(n=10) and 3 years old group(n=10).The expressions of HIF-1α,VEGF and TGF-β1 in the above samples were detected using immunohistochemical staining.Results:The expressions of HIF-1α,VEGF and TGF-β1 were negative or slight in the 2 kinds of tissue for 3 years old group.The expressions of HIF-1α,VEGF and TGF-β1 in the megaureter ending tissue for 1 year old group and 1~3 years old group were significantly higher than those in control tissue(HIF-1α:Z=3.593 and 3.721;VEGF:Z=3.554 and 3.465;TGF-β1:Z=3.554 and 3.552,all P0.001).Conclusion:Alteration of the expressions of HIF-1α,VEGF and TGF-β1 may participate in the pathogenesis of primary megaureter.
