摘要
目的:探讨缺氧条件下肾癌786-0细胞体外黏附和迁移能力的变化。方法:将786-0细胞分别置于正常氧(37℃体积分数为5%CO2和21%O2饱和度,正常组)和缺氧(37℃体积分数为10%O2、5%CO2及85%N2,缺氧组)环境中培养,采用MTT法和Boyden小室法检测2组细胞的黏附能力和侵袭能力。结果:正常组786-0细胞的黏附率为(71.6±6.1)%,与缺氧组的(84.2±4.4)%相比,差异有统计学意义(t=3.748,P=0.006)。缺氧组786-0细胞的穿膜细胞数为(48±6),高于正常组的(28±5)(t=5.722,P=0.001)。结论:缺氧条件下肾癌786-0细胞的黏附和迁移能力增强。
Aim:To study the effects of hypoxia on adhesion and migration of 786-0 renal carcinoma cell in vitro.Methods:The hypoxic condition was the mixed gas of 10%O2,5% CO2,and 85%N2.786-0 cells cultured in hypoxic condition was the hypoxia group,meanwhile cells into 5% CO2,37 ℃ incubator was the control group.The adhesiveness and migration of 786-0 cells in vitro were detected using MTT and Boyden method,respectively.Results:The adhesive rate of 786-0 cells in hypoxia groupt was (84.2±4.4)%,higher than that of control group (71.6±6.1)%(t=3.748,P=0.006).There was significant difference in transwell cells number between hypoxia group(48±6) and control group (28±5)(t=5.722,P=0.001).Conclusion:The adhesiveness and migration ability of 786-0 cells enhance in hypoxic condition.
