摘要
目的:观察常见不同证候荷瘤小鼠下丘脑基因表达的特征。方法:采用H22荷瘤小鼠及其标准化四诊与辨证方法,以及GeneChip Mouse Exon 1.0 ST Array等技术,检测荷瘤小鼠早期邪毒壅盛证和气虚证、中期阳气虚证、中晚期气阴阳虚证等4个证候及正常小鼠下丘脑基因的表达,筛选出不同证候一致上调和下调的基因。结果:荷瘤小鼠一致上调的基因有Cd74、Ctxn1、Lphn1、Npcd、Syn1、Nfix、Nr1d1、C530028O21Rik、Mast3、Nt5dc3、Ttyh3、Dusp8,Bat2、Laptm5、Ptms、Sez6、Atn1、Syt3、Eif5a、Eif3s5、Ap1g2、Ap2a1、Lcn2、S100a8、BC033915、P140、LOC620583共27个;一致下调的基因有Hrsp12、Acta2、LOC675606共3个。结论:肿瘤发生后,H22荷瘤小鼠下丘脑存在大量基因表达的差异,其中部分与不同证候有关。
Objective:To reveal the character of gene expression in hypothalamus of H22 tumour mice with different syndromes.Methods:By the quantitative four diagnosis and syndrome differentiation methods and GeneChip Mouse Exon 1.0 ST Array,hypothalamus gene expression about normal mice,poisonous pathogenic factors syndrome and asthenia of qi syndrome on earlier period,asthenia of yang and qi syndrome on intermediate stage,asthenia of qi and yin and yang syndrome on advanced stage with H22 tumour mice was observed.Genes expressed highly and differentially distinguished were analyzed on the paper.Up or down-regulated coincident genes In different syndromes were screened out.Results:27 genes were up-regulated coincident such as Cd74,Ctxn1,Lphn1,Npcd,Syn1,Nfix,Nr1d1,C530028O21Rik,Mast3,Nt5dc3,Ttyh3,Dusp8,Bat2,Laptm5,Ptms,Sez6,Atn1,Syt3,Eif5a,Eif3s5,Ap1g2,Ap2a1,Lcn2,S100a8,BC033915,P140,LOC620583.Then 3 genes were down-regulated coincident such as Hrsp12,Acta2,LOC675606.Conclusion: After the tumorigenesis,gene expression change was distinguished in hypothalamus of H22 tumour mice.Some differentially expressed genes could be related to different symptoms and signs.
