摘要
目的:探讨白细胞介素-10(IL-10)基因启动子区592 C/A基因多态性与复发性流产(RSA)的关系。方法:应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)及琼脂糖凝胶电泳法检测无血缘关系的298例浙江地区汉族人IL-10启动子区592 C/A基因多态性,其中142例RSA患者(RSA组)和156例健康志愿者(正常对照组)。结果:在RSA组和正常对照组中IL-10592 C/A基因中C等位基因的分布频率分别为40.1%、51.0%;A等位基因的分布频率分别为59.9%、49.0%;CC基因型频率为17.6%、23.7%;CA基因型为45.1%、54.5%;AA型为37.3%、21.2%。两组人群的基因型和等位基因频率表达比较,差异有统计学意义(P<0.05)。RSA组中AA基因型血清IL-10水平4.25±1.32 pg/ml低于CC基因型血清IL-10水平8.15±2.01 pg/ml,差异有高度统计学意义(P<0.001)。结论:IL-10基因启动子区592C/A基因多态性可能与RSA发病有关,A等位基因的出现可能会减少血清IL-10的分泌。
Objective.To investigate the association of interleukin-10 592C/A gene polymorphism with recurrent spontaneous abortion (RSA). Methods.592C/A gene as the initiator gene of interleukin-10 its gene polymorphism was determined by polymerase chain reaction-restriction fragment length polymorphisms assay (PCR-RFLP) in 298 unrelated women of Han population (including 142 women with RSA and 156 women as health control). Results. The allele frequency of A in RSA group was higher than that in control group (59.9% vs 49.0% ). There were significant differences of CC,CA, AA and C allele gene frequencies of IL- 10 592 C/A gene between the two groups (P〈0.05). The serum IL-10 level for AA genotype in RSA group was lower than that for CC genotype (4.25 ±1.32 pg/ml vs 8.15 ± 2.01 pg/ml), the difference was significant( P 〈0.001 ). What's more, the serum levels of IL-10 for AA, CC, CA genotype were significant different when they were compared respectively(P 〈 0. 001 ). Conclusions.The interleukin-10 592 C/A gene polymorphism may involve into the pathogenesis of RSA, the occurrence of A allele may cause the reduction of serum IL-10 secretion.
