摘要
目的观察人肝再生增强因子对IL-10分泌的影响。方法用梯度离心法分离正常人PBMC,分成正常组、hALR组、ConA刺激组和ConA+hALR组。培养16h后,用MTT法观察细胞增殖情况,双抗夹心酶联法检测上清中细胞因子IL-2,IL-10水平。结果 ALR显著抑制ConA对细胞的增殖作用的同时,明显促进细胞因子IL-10分泌和显著抑制细胞因子IL-2分泌。结论 ALR可能通过分泌IL-10和抑制IL-2分泌而发挥免疫抑制作用。
To observe the effect of human augmenter of liver regeneration (hALR) on IL-10 secretion,PBMCs were isolated from healthy volunteers by gradient centrifugation and divided into normal,ALR,ConA and ALR+ConA groups. The proliferation of PBMCs and the levels of IL-2 and IL-10 in the supernatant were detected by MTT and Double-antibody Sandwich Enzyme-linked Immunosorbent Assay respectively when cells were cultured for 16 hours. Correlation between IL-2 and IL-10 was analyzed. As a result,ALR promoted the PBMCs to secret cytokine IL-10,while inhibited the secretion of the cytokines IL-2 and significantly inhibited the ConA positive effect on cell proliferation. In a conclusion,hALR plays immunosuppressive effects possibly through promoting IL-10 secret and inhibiting IL-2 secretion.
