抗氧化剂N-乙酰半胱氨酸对巨噬细胞产生IL-12及IL-23的不同作用 被引量:3

摘要 目的观察N-乙酰半胱氨酸(NAC)对巨噬细胞产生白介素-12(IL-12)及白介素-23(IL-23)的作用。方法 IL-12p35,p40,IL-23p19及IL-17的mRNA表达采用RT-PCR进行检测。IL-12p40,IL-12p70,IL-23及IFN-γ的产生使用ELISA进行测定。结果小鼠巨噬细胞经过NAC刺激6h后IL-12p35和p40的mRNA表达增强,而NAC刺激24h后p35的mRNA表达和IL-12p70的产生被减弱。另一方面,NAC增进了IL-23p19的mRNA表达和IL-23产生。此外,在小鼠脾细胞中,NAC增强了IL-23p19mRNA表达而抑制了IFN-γ的产生。NAC刺激后的CD11b+细胞能够促进T细胞中IL-17mRNA的表达。结论 NAC抑制巨噬细胞产生IL-12而促进巨噬细胞产生IL-23,从而促进Th17免疫反应。 We aim to study the effects of N-acetylcysteine (NAC) on IL-12 and IL-23 expression in macrophages. In this study,we employed RT-PCR and ELISA assays,and which revealed that NAC enhanced IL-12p40 and p35 mRNA expressions at the 6th hour,but inhibited p35 mRNA and IL-12p70 production at the 24th hour after stimulation in macrophages. On the other hand,IL-23p19 mRNA expression and IL-23 production was enhanced by NAC. In addition,IL-17 mRNA was up-regulated by NAC but IFN-γ production in spleen cells was inhibited by NAC. And NAC-treated CD11b+ cells promoted the expression of IL-17 mRNA in T cells,which indicated the biologic IL-23 production. Taken together,these results suggest that NAC differently regulates IL-12 and IL-23 production and preferentially activates Th17 rather than Th1 immune responses.
