脉络膜撕脱伤后碱性成纤维生长因子表达的变化

摘要 目的通过建立兔眼脉络膜撕脱伤模型,观察伤后不同时间点碱性成纤维细胞生长因子(bF—GF)表达的动态变化,探讨脉络膜撕脱伤后bFGF在脉络膜增殖修复中的作用。方法采用固相夹心法酶联免疫吸附实验(ELISA)法检测兔眼脉络膜撕脱伤模型玻璃体中bFGF的含量;用HE染色光镜下观察伤后脉络膜、视网膜组织病理变化;用SP法对伤后脉络膜、视网膜做免疫组织化学染色,染色结果行图像分析。结果玻璃体中bFGF在450nm处平均OD值及脉络膜、视网膜组织bFGF表达平均光密度值均出现先增高又下降的动态变化,伤后第7天达到高峰,伤后第14天有所回落,但仍维持较高水平,伤后第28天明显下降,伤后第7天与伤后第1天、第3天、第28天相比差异有统计学意义(P〈0.05)。结论bFGF是脉络膜撕脱伤后脉络膜增殖、纤维化、瘢痕化过程中的一重要因素,加重了外伤性增殖性玻璃体视网膜病变(PVR)的发生。 Objective To explore the dynamic expression of basic fibroblast growth factor( bFGF)in rabbit choroidal tear trauma and to explore the mitogenic and differentiation properties of bFGF at tissue repairing. Methods Expression of bFGF in vitreous was detected by enzyme-linked immunosorbnent assay ; and the expression of hFGF in retina and choroid was detected by immunohistochemical analysis ; and the histopathologic changes of choroid and retina was observed by hematoxylin( HE). Results After establishing the animal mode, the expression of bFGF in vitreous,retina and choroid was first upregulated and then down-regulated. The peak value was found in the 7d group, which significantly differed from the values in the ld group,3d group and 28d group. Conclusion bFGF contributes to the proliferation,fibrosis and cicatrisation in choroidal tear trauma and promotes the traumatic proliferative vitreoretinopathy ( PVR ). This result give guidance to choose time of vitectomy after choroidal tear trauma.
