摘要
目的:探讨灵芝三萜对口腔黏膜癌的防治作用及其作用机理。方法:建立金黄地鼠颊囊动态癌变模型60只,分2组:实验组(灵芝三萜组)和对照组(未服用灵芝三萜组),每组各30只,对所取标本进行病理检验,并采用免疫组化技术检测bcl-2和survivin在两组动物模型中的表达变化。结果:病理结果显示随DMBA涂药时间的延长,两组动物模型的病变呈加重趋势(P<0.01),第6、9、12周实验组的病变程度明显低于对照组(P<0.05);从总体上看两组颊囊癌变过程中不同组织学发生情况有显著差异(P<0.01)。Bcl-2在上皮异常增生阶段、survivin在上皮单纯增生和异常增生阶段,实验组的表达强度明显低于对照组。结论:灵芝三萜对口腔黏膜癌具有抑制作用,其作用机制可能与凋亡调节基因survivin、bcl-2的下调有关。
Objective:To explore the preventive effect of triterpenoids isolated from Ganoderma lucidum on cancerization of the oral mucosa and the underlying mechanism.Methods:60hamster cheek pouch dynamic cancer animal models were established and divided into two groups:experimental group(Ganoderma triterpenoids treated group) and control group(without Ganoderma triterpenoids treated group).The difference of pathological changes between the two groups were examined under a light microscope at the period of 6,9and 12weeks after operation,and the expression of bcl-2and survivin in each group was detected by immunohistochemistry.Results:The extent of canceration in experimental group was significantly lower than that of control group at the period of 6,9and 12weeks after operation(P0.05).In the whole dynamic carcinogenesis observation period,the occurrence of two groups'histological difference was significant(P0.01).At period of epithelial dysplasia,the expression of bcl-2in experimental group was significantly lower than that of control group.At period of epithelial dysplasia and epithelial proliferation,the expression of survivin in experimental group was significantly lower than that of control group.Conclusion:Ganoderma triterpenoids may inhibit cancerization of the oral mucosa and this inhibitive effect may be related to down-regulating expression of bcl-2and survivin.It may be used for treatment of oral squamous cell carcinoma.
