摘要
目的:构建MAGE1和MAGE3融合表达载体,并获得抗血清。方法:采用分步交叉亚克隆方法将两个不同片段连接于表达载体,分组对照优化表达条件,用SDS-PAGE和Western-blot检测表达与纯化结果,将纯化蛋白免疫动物后用琼脂双扩和ELISA方法检测抗血清效价。结果:成功构建了MAGE1-MAGE3表达载体并获得纯化蛋白;免疫动物后获得高效价的抗血清。结论:得到纯度高,变性度低的融合蛋白。
Objective:To construct combinant MAGE1 and MAGE3 expression vector and acquirement of rabbit anti-serum of combinant protein.Methods:To construct expression vector by truncated sub-clone and cross clone; verified by SDS-PAGE and Western-blot; agar precipitation and ELISA detected the titer of anti-serum.Results:Successfully constructed the expression vector on pET-28a.Conclusion:Acquired the optimal expression conditions and combinant protein,and obtained high titer anti-serum.
