摘要
目的:探讨生长抑素类似物奥曲肽能否提高化疗药物对于人肺癌细胞株A549敏感性。方法:以体外培养的人肺癌细胞株A549为靶细胞,运用四氮唑盐法(MTT法)观察奥曲肽单独作用及与化疗药物顺铂共同作用对于A549生长的影响,并以流式细胞仪分析其对细胞周期的影响。结果:MTT法结果显示奥曲肽抑制A549的生长,浓度0.098mg/L-25mg/L范围内,呈剂量依赖性,与顺铂共同作用后,能够增强顺铂对于A549的敏感性。流式细胞仪测定结果显示:奥曲肽作用后,G0/G1期细胞比例增加,G2/M期细胞比例减少,二者共同作用后,此趋势更加明显。结论:奥曲肽能够抑制体外培养的人肺癌细胞株A549增殖,且与顺铂有协同作用,其机制可能是发生G0/G1期阻滞。
Objective:To explore whether octreotide, as somatostatin analogue,could enchance the chemotherapeutic drug sensitivity in human non-small cell lung cancer cells A549.Methods:A human non-small cell lung cancer cells line (A549) was cultured,octreotide and chemotherapeutic drug cisplatin were added.MTT value was tested by colorimetry to evaluate the cell activity of octreotide,cisplatin and the both respectively,flow cytometry (FCM) was used to investigate the influence of cell cycle.Results:Octreotide could inhibit the proliferation of A549 cells in dose-dependent manner especially in the concentration range in 0.098-25 mg /L,the inhibition rate of cisplatin + octreotide group was significantly higher than that of cisplatin group and octreotide group,respectively.The cell cycle analysis showed that the percentage of G0/G1 phase cells increased and G2/M phase cells decreased after treatment with octreotide and there was significant tendency after treatment with combination of octreotide and cisplatin.Conclusion:Octreotide could inhibit the proliferation of A549 cells in vitro,and it enhance the cytotoxity of cisplatin,the mechanism may be related to its preventing the cell cycle in G0/G1 phase.
