槲皮素干预实验性慢性肾衰大鼠肾脏一氧化氮超氧化物岐化酶与过氧化脂质变化 被引量:4

Effects of Quercetin on Renal Superoxide Dismutase and Lipid Peroxides Changes in Rats with Experimental Chronic Renal Failure

摘要 目的探讨槲皮素干预实验性慢性肾衰大鼠对肾脏一氧化氮、超氧化物岐化酶和过氧化脂质的影响。方法给实验大鼠饲以含腺嘌呤饲料2个月造成慢性肾衰;干预组经口给予槲皮素100mg/(kg.d)×42d;按试剂盒指定方法测定肾组织一氧化氮、超氧化物岐化酶和过氧化脂质。结果与肾衰对照组比较,干预组大鼠肾脏组织一氧化氮含量显著升高,平均每只肾(570±193)和(263±123)nmol,超氧化物岐化酶活性显著升高,平均每只肾(7.68±2.98)和(3.26±2.63)Ku,过氧化脂质量亦显著增加,平均每只肾(103.3±16.1)和(39.2±17.5)nmol,肾组织细胞核因子-κBP65丢失情况明显减轻。结论槲皮素干预可显著提高实验性慢性肾衰大鼠肾脏一氧化氮、超氧化物岐化酶和过氧化脂质,提示其可保护肾脏细胞及抗氧化,减轻细胞损伤。 Objective To observe the effect of quercetin on nitrogen oxide,superoxide dismutase and lipid peroxides in rats with experimental chronic renal failure.Methods The rats in the control and treatment group were fed with 0.75% adenine for 2 months to induce chronic renal failure.Quercetin was administered orally at a dose of 100 mg/(kg·d)for 42 days to the rats in treatment group,beginning at the 15th day.Nitrogen oxide,superoxide dismutase and lipid peroxides in renal tissues of the experimental rats were determined using the methods designed in the bought kits of assaying agents.The Nuolear Factor-κB in the renal cells were assayed with a immunochemical assay.Results When compared with the control group,the level of nitrogen oxide in renal tissues in the treated group of rats was increased(570±193)nmol vs(263±163)nmol,so was the renal superoxide dismutasewere(7.68±2.98),(3.26±2.63)Ku and the renal lipid peroxides(103.3±16.1)nmol vs(39.2±17.5)nmol(P0.001).The loss of Nuolear Factor-κB P65 in renal cells in the treated group of rats was less serious than that in the control group.Conclusion The results showed that qercetin significantly elevated nitrogen oxide,superoxide dismutase and lipid peroxides levels in renal tissues of chronic renal failure rats suggestny its effect of anti-oxidation and remeidying injuries in cells of the kidney of treated rats.
